Mike O'Brien studied film video at the University of Michigan and was the founder and editor of The Anti-Daily, a monthly comedy publication. He received his diploma in 1999. In Chicago, he performed at iO Chicago and The Second City. He began his training at Chicago's Second City, eventually joining the company's mainstage, where he stayed for over 14 years.
He and Mike O'Brien co-created the online series 7 Minutes in Heaven, in which he uncomfortably converses with someone in a closet for a few minutes before attempting to kiss them. O'Brien auditioned for a role as a cast member in 2005 and 2009, the latter of which led to his being hired as a writer for the 2009–10 season.
